MimiVax, a Brain Tumor Investment Fund™ portfolio company, and Roswell Park Comprehensive Cancer Center have reported results from SURVIVE (NCT05163080). The trial is a phase 2b randomized, double-blind, placebo-controlled study of the company’s investigational vaccine, SurVaxM, in people with newly diagnosed glioblastoma (GBM), with 233 evaluable patients at 11 U.S. cancer centers. According to the company, median overall survival, the trial’s primary endpoint, was 22.8 months with SurVaxM and 20.3 months with placebo (HR 0.75; p=0.076), which did not reach statistical significance. In a prespecified subgroup of patients 65 and under, median overall survival was 24.2 vs. 20.2 months (HR 0.64; p=0.019), a statistically significant difference.
SurVaxM targets survivin, a protein present in most glioblastomas that helps cancer cells avoid programmed cell death. The vaccine is given by injection under the skin and is designed to prompt the immune system to recognize and attack tumor cells that express survivin. In SURVIVE, it was given alongside standard of care. The company reported that SurVaxM was well tolerated, with mostly mild to moderate injection-site reactions. MimiVax plans to review the data with the FDA.
